We show that in the presence of TGF beta signalling , Smad 2 interacts through its proline rich PPXY motif with the tryptophan rich WW domains of Smurf 2 , a recently identified E 3 ubiquitin ligases . ^^^ TGF beta also induces the association of Smurf 2 with the transcriptional co repressor SnoN and we show that Smad 2 can function to mediate this interaction . ^^^

Smurf 2 ( Smad ubiquitination regulatory factor 2 ) was identified as the Smad ubiquitin ligase that induces the ubiquitination and degradation of Smad 2 . ^^^ This study was undertaken to elucidate the relationships between Smurf 2 expression and the clinicopathological characteristics of patients with esophageal squamous cell carcinoma ( SCC ) and the correlation between Smurf 2 and Smad 2 expression . ^^^ We also found an inverse correlation between the expression of Smurf 2 and Smad 2 . ^^^ We demonstrated that high level expression of Smurf 2 appears to correlate with tumor development and poor prognosis in patients with esophageal SCC and that alteration of Smad 2 expression in the TGF beta signaling pathway may be induced by enhancement of Smad 2 degradation mediated by high level expression of Smurf2 . . ^^^

Since Aza unmasked responsiveness of ACs to BMP 2 and TGF beta , we examined the effect of Aza treatment on signaling via these pathways by assessing the expression of the TGF beta Smads ( 2 and 3 ) , the BMP 2 Smads ( 1 and 5 ) , and the Smad 2 and 3 degrading ubiquitin E 3 ligase Smurf 2 . ^^^ Aza treated ACs displayed less Smad 2 and 3 and increased Smad 1 , 5 , and Smurf 2 protein and showed a loss of TGF beta signaling on the P3TP luciferase reporter . ^^^ Suggesting that Aza induction of Smurf 2 may be responsible for the loss of Smad 2 and 3 protein via this pathway , immunoprecipitation and metabolic labeling experiments confirmed that Aza accelerated the ubiquitination and degradation of these targets . ^^^ Overall , Aza treated ACs represent a novel model for the study of mechanisms that regulate maturational potential of articular cartilage , with the data suggesting that maturation of these cells may be due to up regulation of Smad 1 and 5 coupled with a Smurf 2 dependent degradation of Smad 2 and 3 and loss of TGF beta signaling . . ^^^
