We identified the homeodomain protein TGIF as a Smad 2 binding protein and a repressor of transcription . ^^^ Thus , upon entering the nucleus , a Smad 2 Smad4 complex may interact with coactivators , forming a transcriptional activation complex , or with TGIF and HDACs , forming a transcriptional repressor complex . ^^^

Jun interacts with the corepressor TG interacting factor ( TGIF ) to suppress Smad 2 transcriptional activity . ^^^ This inhibitory activity is mediated through the transcriptional factor c Jun , which enhances the association of Smad 2 with the nuclear transcriptional corepressor TG interacting factor ( TGIF ) , thereby interfering with the assembly of Smad 2 and the coactivator p 300 in response to TGF beta signaling . ^^^ Interestingly , c Jun directly binds to the nuclear transcriptional corepressor TGIF and is required for TGIF mediated repression of Smad 2 transcriptional activity . ^^^

The novel E 3 ubiquitin ligase Tiul 1 associates with TGIF to target Smad 2 for degradation . ^^^ Here , we identify Tiul 1 ( for TGIF interacting ubiquitin ligase 1 ) , a novel E 3 ubiquitin ligase that inhibits TGF beta signaling by targeting both the activated receptor and Smad 2 for degradation . ^^^ Tiul 1 can also interact with Smad 2 and the nuclear corepressor TGIF upon activation of TGF beta signaling . ^^^ Like Smad 7 , the steady state levels of TGIF are not affected by Tiul 1 , but the interaction of Tiul 1 with TGIF allows this ubiquitin ligase to target Smad 2 for degradation . ^^^ In addition , silencing of Tiul 1 or TGIF genes by siRNA resulted in suppression of the TGF beta dependent degradation of Smad 2 and an enhancement of TGF beta mediated gene expression . ^^^
