Here , we show that huntingtin copurifies with a single novel 40 kDa protein termed HAP 40 . ^^^ In transfected cell extracts , HAP 40 coimmunoprecipitates with full length huntingtin but not with an N terminal huntingtin fragment . ^^^ Recombinant HAP 40 is cytoplasmic in the presence of huntingtin but is actively targeted to the nucleus in the absence of huntingtin . ^^^ These data indicate that HAP 40 is likely to contribute to the function of normal huntingtin and is a candidate for involvement in the aberrant nuclear localization of mutant huntingtin found in degenerating neurons in Huntington ' s disease . . ^^^

Remarkably , endogenous HAP 40 was up regulated in fibroblasts and brain tissue from human patients affected by Huntington ' s disease ( HD ) as well as in STHdhQ ( 111 ) striatal cells established from a HD mouse model . ^^^ In this study , we have identified Htt associated protein 40 ( HAP 40 ) as a novel effector of the small guanosine triphosphatase Rab 5 , a key regulator of endocytosis . ^^^ HAP 40 mediates the recruitment of Htt by Rab 5 onto early endosomes . ^^^ In revealing an unexpected link between Rab 5 , HAP 40 , and Htt , we uncovered a new mechanism regulating cytoskeleton dependent endosome dynamics and its dysfunction under pathological conditions . . ^^^ Huntingtin HAP 40 complex is a novel Rab 5 effector that regulates early endosome motility and is up regulated in Huntington ' s disease . ^^^
